Title: Human T cell function in healthy and inflamed mucosal tissues

T cell fate decisions in healthy and inflamed human mucosal tissues remain poorly defined despite insight provided from the mouse model system. This includes conventional T cells as well as innate-like T cells such as mucosal-associated invariant T (MAIT) cells. To study these T cell populations in health and disease, we developed an analysis pipeline that allows us define their location (IF) and function (FACS, RNAseq) within human mucosal tissues and interrogate mechanisms that control cell fate decisions in ex vivo assays. Our overall goal is to understand the functional properties of T cells in these healthy vs. inflamed mucosal tissues, but we also provide an example for how we leverage this information to define and test potential therapeutic intervention strategies to decrease function (autoimmunity) or restore function (tumors).
